Yes, it is possible... here's a test I use to test focus & DOF:
Set up camera on tripod outdoors on a flat surface, e.g. concrete, in good light. Lay a tape measure along the ground, pointing directly outward along the lens axis, and focus on a known spot (use manual focus). The angle between the camera & tape should be low, but great enough that the markings are distinguishable.
Focus on the chosen mark at full wide aperture (center the mark in the DOF), trigger the shutter and review the results to see if the DOF is centered on the mark. Repeat the test at increasing F stops (don't move focus!). Repeat above procedure with other zoom settings if required.
