In short, yes, you would, barring mechanical incompatibility. With the 2.0x extender, for instance, you'd wind up with a 100mm f/5.0 lens with double the maximum magnification. Also FWIW, if the 50/2.5 will not mount on the extender due to mechanical interference, it's possible to mount an extension tube between them to eliminate the interference. You then lose infinity focus, and gain a little more magnification.
You may or may not be satisfied with the ultimate image quality. A simpler strategy is just to use 50mm of extension tubes, which will have approximately the same effect on magnification.
